Danny Fenton[]
Boyfriend/Best Friend/Husband (in a Potential Future) Danny is Sam's primary love interest. However, they first started off as best friends and have known each other since at least the second grade. When Danny was made half-ghost and doubts himself, Sam usually encouraged him to embrace his ghost half because it was what made him special. Since then, Sam has helped him fight off ghost supervillains and kept his identity a secret. It is shown they are both jealous wherever the other has a crush on someone else; Danny felt jealous when she began a brief romantic relationship with exchange student Gregor and Sam feels it vice versa with Danny's crushes on Paulina and Valerie Gray.However, they are initially unsure about their romantic feelings for each other, and it isn't until they realize their mutual interest towards each other at the end of the series. Before they have to save the earth, Danny and Sam enter a passionate kiss that begins their relationship.
After years of people pointing out their feelings for each other, they begin a relationship as boyfriend and girlfriend, which would continue well after the Disasteroid crisis, though it would face many outside hinderances.By the time of their college years, Danny and Sam often struggle to find alone time together, due to the hordes of paparazzi and fans that are always after Danny Phantom. This causes them much frustration, forcing them to go on invisible dates, in which Danny would have to make them both invisible in order to hide from the crowds. Despite this, they still continue to maintain a healthy and stable relationship while battling ghosts on Team Phantom ten years following Danny acquiring his ghost powers.

Tucker Foley (Best Friend)[]
Tucker is one of Sam's best friends, along with Danny Fenton. They are bonded through being the only ones aware of his secret as Danny Phantom and supporting him. However, Sam does not like it when Tucker is a bad influence on Danny or nearly gets him killed. (Mystery Meat). Despite considering him an annoyance, as she expressed happiness that Danny used his Intangiblity power to make Tucker fall through the lab's floor when helping him train in "Maternal Instincts," she enjoys his company. This is proven in the episode "Lucky Love," where they both missed hanging out with each other (and Danny) after being forced to hang out with Star and Kwan separately. When Kwan and Star got back together, Tucker and Sam immediately rushed out of the Nasty Burger restaurant and were glad to be rid of the popular kids. They have also partnered up on certain projects, namely the one where they partnered up on taking care of a sack of flour in "Life Lessons" and becoming his personal trainer so he could pass his physical examination in "Micro Management'. In "13," one point when Tucker was considered bad luck/Sam convinced him to go goth like her and to embrace his new image. However, Tucker decides to he himself despite Sam's help. She later broke up with Gregor because he insulted Tucker, showing she cares more about her longtime friends than a relationship with a guy she hardly knew. They also disapprove of Danny's friendship with Valerie because they know that she will "shoot first and ask questions never" and seem to become overprotective of him when he joins her. When Valerie showed interest in Danny, the two sic Nate on her, so she ran away. Along with Danny, she and Tucker work well with each other on their missions and remain the first individuals to know Danny's secrets.
Dorathea[]
The ghost princess/dragon shapeshifter was one of Sam's greatest enemies. Later, Sam helped Dorathea realize that her brother did nothing but make her feel worthless and ashamed for who she was. She showed her to be just as strong as her brother and helped her defeat Aragon once and for all.
Jazz Fenton (Close Friend/Sister-in-law)[]
At the start of the series, Sam and Jazz little to no interaction with each other at the start of the series but were on good terms with each other. While they were not close, Sam was skeptical of Danny’s claims of Jazz calling him a loser, noting that it “doesn’t sound like Jazz”. This implies that they had a fairly good aquaintance with each other, with Sam even reassuring Jazz not to worry about Danny after he started acting strangely. Since Sam has been best friends with Danny since the second grade, she has probably known Jazz since then.
After Jazz made it known that she learned about Danny's secret back with the incident with Penelope Spectra, she joined Sam and Tucker as members of Team Phantom. Initially, Sam found Jazz unfit to become a ghost hunter like them, with this being made clearer in the season 3 episode "Girls' Night out." However, the two girls bond over the course of the episode when dealing with Spectra, Kitty and Ember. Since they learn that they should accept each other's own opinions and ideas, Jazz and Sam effectivity become close friends and now learn to support each other as well as Danny when he is dealing with his superpowers. This is shown in the episode ”Frightmare” when they both easily agree on how to help Danny defeat Nocturn. Despite becoming closer friends, Jazz (in contrast to her parents) seemed to be oblivious to Sam and Danny’s growing feelings to each other, being visibly surprised by their tender moment before entering Nocturn’s dream. When the world was saved from the Disasteroid, Sam and Jazz embraced each other in a passionate hug of relief and celebration.
Jack and Maddie Fenton[]
Unlike Jeremy and Pamela's relationships with Danny and Tucker, Sam actually gets along with Danny's parents. Both Jack and Maddie appear to treat both Sam and Tucker like family. In "Girls' Night Out," Maddie worked relatively well with Sam (and Jazz) in capturing Kitty, Spectra, and Ember, and Jack is aware of Sam and Danny's feelings for each other.

Jeremy and Pamela Manson (Parents)[]
Jeremy and Pamela Manson are Sam's parents; however, they don't see eye to eye because of their conflicting personalities. They disapprove of Sam's gothic appearance and her friendships with Danny, even getting a restraining order against him, or with Tucker. They mostly don't like Danny because of his parents' ghost hunting obsessions.
Despite this, Sam loves her parents despite them getting on her nerves.
Ida Manson (Grandmother)[]
Ida is Sam's grandmother and the one person in her family who understands her and loves her for who she is. She affectionately calls Sam "Bubeleh," which is a term of endearment, like "sweetie" or "honey."" In "Control Freaks," Ida let Sam sneak out of the house to save Danny from Freakshow's control after her parents forbade her from being friends with him, which Sam responded by kissing her grandmother on the cheek before leaving.
In the future, Ida is the great-grandmother of Danny and Sam's children.
Enemies[]
Paulina Sanchez[]
Sam dislikes Paulina Sanchez forever. A variety of reasons: she finds her attitude stuck up, arrogant and pompous. However, her dislike for Paulina is because Sam is partially jealous of her because Danny, Sam's own crush and best friend, romantically likes Paulina. When Sam was wearing Dorathea's amulet, she grew agitated when Sam discovered Paulina only went out with Danny to make her jealous and that she was just using him. She transformed into a dragon.
In a twist of irony, both girls are from rich families; however, Sam knows not to advertise her being rich because she wants friends who are true to her. Paulina feels the same way about Sam.

Kitty[]
Like Ember and Penelope, Kitty appears to be becoming one of Sam's personal enemies. Since Danny is the one who fights the ghost opponent for the episode while Sam usually helps fight them, she rarely interacted with Kitty. However, this changed in Season 3's episode, "Girls' Night Out!" Because of Danny's absence, Sam, Maddie, and Jazz have to fight the three ghost girls; this makes Kitty (along with Penelope and Ember) one of her personal opponents.

Valerie Gray (Former Rival)[]
Sam admits she cannot hate Valerie because they hardly know her, but sometimes, reminds the former popular girl that she used to treat Sam and her friends, Tucker and Danny, poorly.
Mostly, Sam does not like Valerie because she poses as a threat to Danny and knows she will hurt him if Valerie ever finds out the truth. However, it is implied that Sam gets jealous of Valerie because Danny likes her as well. Some of the other reasons are that Sam and Valerie each had romantic feelings for Danny, and Valerie acknowledged Sam liked him but warned her that she better make a move before someone else did. During Valerie's short-lived romance with Danny, Sam spied on them in their dates, and when she approached Valerie, Sam gave her a fake blessing. In return, Valerie, is genuinely touched by this and hugs Sam for her understanding (who is mildly uncomfortable with this), which also shows that Valerie wants to have a possible friendship with Sam and genuine understanding with her because of their shared feelings for Danny.
